Not sure if these are my definite top 5, but they're the ones who came to mind when I saw the thread title.

Margo Channing (Bette Davis) - All About Eve
Eve Harrington (Anne Baxter) - All About Eve
Beatrice (Emma Thompson) - Much Ado About Nothing
The Joker (Heath Ledger) - The Dark Knight
Ferris Bueller (Matthew Broderick) - Ferris Bueller's Day Off*

*I love all three of them really, it's hard to choose between them but since he's the star.

James Garner as Hendley in the Great Escape is brilliant, but then Great Escape & Magnificent Seven were full of terrific characters.

Leslie Nielsen as the doctor in Airplane would have to be in there, Rowan Atkinson as the vicar in Four Weddings, throw in Steve McQueen/Hiltz and Pele for his goal in Escape to Victory for my 5.

Harsh on the knight (Cleese) who gets enhiliated at the start of the Holy Grail with his "flesh wound" who just misses out.

James Bond. Connery, Moore, Dalton, Brosnan. Certainly NOT Craig, whose films have meant I hate current Bond. They're not Bond movies!

Chucky. The killer doll whose, simultaneously cute and disturbing.

Michael Myers. The mysterious killer from the original Halloween series.

    Ajax. The Warriors.

Peter from the original Dawn of the Dead. Somehow charismatic while blasting zombies!

Mentions too, to Kevin McCalister (Home Alone) and Indiana Jones.
