As for Stanley Holloway I've just realised he was one of the crooks in The Lavender Hill Mob which I suppose makes him a villain.
I don't think Esme Cannon who specialised in playing dotty old ladies ever played a villain

He was essentially the villain of the piece in Dreamgirls. Not a villain in the true sense of the word, but certainly the horrible guy in the movie!
Not sure about the others, but Dan Aykroyd played a ruthless assassin in Grosse Pointe Blank.
There's loads of villans who's never been actors. Too many to mention really.
I'd personally class him as a villain in The Nutty Professor. But, then, he had various roles in that.
