Will she still be a grandfather?

No she would become a grandmother.

She would still be a grandfather. Hayley from Coronation Street is still a father despite being a woman now.

The Doctor should be played by an American actor because there's no reason not to and there's nothing saying he has to be British all the time. He's even regenerated with different accents before.
