Although The Wire is the best (with Deadwood and The Sopranos not far behind), Buffy the Vampire Slayer will always be my favourite.
And as for comedy, The Simpsons and Seinfeld seem pretty insurmountable. Both had 9 really good seasons, and I'm not sure I can see any other show matching that. Frasier didn't quite manage it, Curb Your Enthusiasm might.
