How long from request and being able to unlock with itunes does it take on O2 pay as you go?

Within a few days usually. If you used the online form you'll get an email when it's done.

If it's running iOS 5 or above then you don't need to unlock it through iTunes. As long as you have an internet connection on the phone it will unlock when a different network's SIM is installed.
