The Boy Is Mine - Brandy & Monica.

20 years later, this duet still hasn't been surpassed.
It really was like catching lightning in a bottle by pairing these two together with such a killer song.

Paul McCartney and Stevie Wonder - Ebony and Ivory (bliddy awful song though)

I bet not many people realise that Elton John & Cliff Richard did a fabulous duet together which charted during Christmas 1986 but only just missed the top forty. It was called Slow Rivers.

Others worth a mention are;

You Are Everything by Diana Ross & Marvin Gaye - No5 in April 1974.

Something's Gotten Hold Of My Heart by Marc Almond & Gene Pitney - No1 in 1989. Weird combo worked.

With You I'm Born Again by Billy Preston & Syreeta - No2 in January 1980. Fabulous love song.

Enough Is Enough (No More Tears) by Donna Summer & Barbra Streisand - No3 in December 1979.

On My Own by Patti LaBelle & Michael McDonald - No2 in May 1986. Kept off the top for 3 weeks by Chicken Song!

Great song. Guilty by Streisand and Gibb as mentioned upthread.

Some others
She Means Nothing To Me - Cliff Richard and Phil Everly
You Don't Bring Me Flowers - Barbra Streisand and Neil Diamond

I'm not normally a fan of opera style songs but I like this one
Time to Say Goodbye - Sara Brightman snd Andrea Bocelli

I take this thread back now - one of the very few songs I can remember that sent a shiver down my spine when I heard it, fantastic song.

Will Young and Gareth Gates - The Long And Winding Road, is massively underrated.

Presumably, the OP is referring to two singers who don't normally record together doing so as a one-off venture (single or album), which therefore prohibits countless wonderful tracks by regular duos (most notably Simon and Garfunkel, the Everly Brothers and the Righteous Brothers) from being included.

Here are some great tracks, which fall into the category of what I perceive the OP requires:-

Something Stupid - Frank and Nancy Sinatra
Some Velvet Morning - Nancy Sinatra and Lee Hazelwood
Passing Strangers - Billy Eckstine and Sarah Vaughan
Mockingbird - Carly Simon and James Taylor

Syreeta and Billy Preston 'With You I'm Born Again' - absolutely beautiful, makes me cry whenever I listen to it.

Ooh thank you - you've given me an excuse to post this - James and Carly singing You Can Close Your Eyes. OK, so it doesn't fit the thread as it wasn't a chart single, but I love it - James sings the melody and Carly sings harmony underneath the melody. Gorgeous.

Doesn't fall in to the OP's definition of a duet but the KLF feat. Tammy Wynette's Justified And Ancient is a great song.

R & J Stone - We Do It.

A No5 hit in January 1976. They were a married couple, Russell and Joanne. The female singer cruelly passed away not long after their only hit of a brain tumour.

The song is well worth a listen;
