A colleague has loaned me a neat little device which allows you to easily measure the signal coming through your aerial cable (it displays the signal for pre-configured channel numbers in dB).
So I had a quick play with it at the weekend, and discovered that the signal coming out of tuner #1 on the DHD4000 is actually boosted by approx 3dB. No need to worry about the second tuner receiving a worse signal than the first - it is actually the other way around!
I also tested the connector which joins the two tuners, and there was no loss across it (in fact I measured a little gain!).

I tested a booster on my signal (only at the STB, so I was also boosting noise - I didn't have the correct connectors handy to attach the booster nearer the masthead), and compared the numbers given by the Thomson. A boost of 10db in the incoming signal, gave an improvement in signal/quality from average 6/7 to average 7/7.
Whilst this won't make much difference (digital signal tends to either work or not work IME), my MuxB reception is currently marginal, and the boost seemed to make the difference to make it more stable - i.e. BBCi worked whereas normally it doesn't.
