If you buy a new Sony Ericsson, it says in the quick start guide/manual that you need to give it 2.5h charge, is this its very first charge?

Yes, modern batteries don't need a long initial charge.

With NiCad batteries they did need an initial long charge. But modern batteries do not. In fact overcharging increases the temperature of the battery which shortens the life. That's why nearly all modern phones have a charging circuit that cuts off the charge to the battery when it is fully charged.
